Product Description

HyperLadder™ 100bp is a molecular weight marker, composed of a restriction digest plus one or more PCR products, especially designed for easy size determination of linear double-stranded DNA fragments on 1% to 2% agarose gels.

The 10 regularly spaced bands, ranging from 100 bp to 1013 bp, with a doublet band at 300/311 bp and higher intensity reference bands at 1013 bp allow for easy identification and orientation of your band at a glance.

This ready-to-use format reduces handling steps and saves time; simply transfer HyperLadder 100bp from the vial to the gel. The concentration of DNA in each of the HyperLadder 100bp bands is provided for optionally determining the approximate mass of DNA in comparably intense sample bands of similar size. The HyperLadder 100bp is also supplied with a 5x loading dye for loading of sample DNA.

HyperLadder 100bp is perfect for size determination in techniques such as confirmation of plasmid construction and PCR products as well as other downstream techniques.

HyperLadder 100bp

What are the optimal conditions for running my ladder?
All HyperLadders and EasyLadders are ready-to-load and require no addition of loading buffer prior to use. In general, the smaller the DNA product, the higher the percentage of Agarose required in the gel. As a guide for optimization we would suggest to use the EasyLadder I with 2% (w/v) agarose gel, Hyperladder 100bp with 2% (w/v) agarose gel, Hyperladder 1kb with 1% (w/v) agarose gel, Hyperladder 50bp with 1.5% (w/v) agarose gel and Hyperladder 25bp with 3.5% (w/v) agarose gel. Please have a look into the related application note for more information about ladder migration in different gel concentrations.
My ladder does not look as it should when I run it, what should I do?
Problems with our HyperLadder and EasyLadder are extremely rare, so please make sure you have carried out each of the following correctly: - Used the correct percentage of agarose in your gel for the specific marker in question. - Used 5 µL of the marker per lane. - Used the correct concentration of stain (e.g. EtBr) to visualise correctly. - Vortexed the marker gently before use. Should problems persist, please contact technical services for further assistance.
Do I need to pre-treat my HyperLadder / EasyLadder prior to loading?
No pre-treatment, such as denaturing, is necessary prior to use.
Do I need to add loading buffer to my Meridian marker to load it onto my gel?
No, our Ladder range is supplied in a ready-to-load format to be directly loaded onto your agarose gel. An additional vial of 5x Loading Buffer is supplied with each of our markers for use with your samples.
Are your ladders suitable for use in Polyacrylamide Gel Electrophoresis (PAGE)?
Our HyperLadder and EasyLadder range is designed for use on Agarose gels and we do not recommend them for PAGE. However, should you wish to use Meridian HyperLadders in this technique, HyperLadder 25bp, our small fragment DNA marker, would be the most appropriate.
We do not wish to stain our gels using EtBr, will other stains still resolve the HyperLadder / EasyLadder correctly?
Other stains, such as SYBR® Green Gel Stain, can be used with our ladders as effectively as EtBr. The fluorescence of the gel as a whole may vary depending on the stain used, however the relative intensities of the marker bands and of the samples run will remain the same, thus the HyperLadder / EasyLadder remains quantitative.
What are the conditions of the gel image in the product insert?
EasyLadder I: gel with 2% (w/v) agarose in TBE HyperLadder 1kb: gel with 1% (w/v) agarose in TBE HyperLadder 100bp: gel with 2% (w/v) agarose in TBE HyperLadder 50bp: gel with 1,5% (w/v) agarose in TBE HyperLadder 25bp: gel with 3,5% (w/v) agarose in TBE For all 5 µL ladder have been applied and visualized using ethidium bromide.
